Dietary copper and zinc have been suggested to have a negative impact on the incidence of diabetes and pancreatic cancer. But the evidence is not conclusive, and this is the first meta-analysis to examine the effects of copper and zinc intake on diabetes and pancreatic cancer. The PubMed, EMBASE, Web of Science, and Scopus databases were systematically searched for prospective cohort and case-control studies reporting on the relation of dietary copper or zinc with the risk of diabetes or pancreatic cancer. Random-effects models were used to examine the summary relative risks (RRs) and 95% confidence intervals (CIs) for the highest versus lowest quantiles of each dietary predictor. At least 4 of the 24 studies examined copper, which was inversely associated with the risk of diabetes (pooled RR, 0.75; 95% CI, 0.55-1.01; I2 = 77%) but not with pancreatic cancer risk (pooled RR, 0.96; 95% CI, 0.71-1.30; I2 = 70%). At least 5 of the 19 studies examined zinc intake, which was inversely associated with the risk of diabetes (pooled RR, 0.77; 95% CI, 0.64-0.93; I2 = 45%) but not pancreatic cancer risk (pooled RR, 0.97; 95% CI, 0.74-1.26; I2 = 75%). The effect estimates for both nutrients were similar. Results were not sensitive to changes in study design, number of cases and adjustment for confounding factors, and the pooled RRs were much higher for copper than for zinc. On the basis of the results of the present meta-analysis, higher copper and zinc intakes do not seem to influence the risk of diabetes and pancreatic cancer.
